2001: A Space Odyssey (1968)
As john-hitchcock mentioned in the discussion, the novel is based on the film, but both were partially inspired by Clarke's short story "The Sentinel". On the other hand, the poll doesn't require the books be written before the movies, although that seems the natural order of things :) In any case, I thought the movie deserves the mention.
